Definitely take a wander along the strip and see the various streetside shows, the Bellagio fountains, and so on.
However, to be honest, our favorite day in Vegas was hiking out at Red Rock Canyon for a couple hours. Some rather scary moments for me edging along some narrow ledges when we got up into the rocks, but it was a day well-spent. Then again, my family all enjoy getting out in the fresh air. Our next vacation after that was Yellowstone National Park.
If it is your first time get a tram pass. Stop at the different casinos for the architecture alone. While you are there register for players cards. They always give you a bunch of free play. There is a ton of free lounge shows and really cool ones at Circus Circus. Check Internet for schedule. Old town Freemont has animated canopy at night. There are a bunch of Buffets that are excellent. Paris buffet and MGM Grand are a couple of my favorites. If you want to get out of Vegas and see something spectacular, go see the Dam.

You must go downtown to the Golden Nugget. The strip is gaudy, glittery plastic and lights; very laid back. The Golden Nugget downtown is marble, brass crystal chandeliers, quiet and dripping with class. You see women in long gowns and dresses, men in suits and tuxes, but also the casually dressed; at least the last time I was there, which admittedly was a number of years ago.

There's also the Atomic Testing Museum, the container park down by Fremont St., the High Roller viewing wheel, plenty of good shows on the strip but they also have the Smith center for the performing arts.

Cirque du Soleil - Zumanity (New York-New York Hotel and Casino) is a once in a lifetime must see!
I get to Las Vegas quite often and agree with what the other posters have said for you to experience.
The previous poster (Californiawilliam) has touched on of Las Vegas' best gems. There are a plethora of Cirque du Soleil shows at the major hotels in town and I would highly recommend you go to one or two. I would strongly suggest "ONE" which is all about Michael Jackson, at one point he even makes an appearance on stage. Definitely a must see!
The other place you have to go is the piano bar in New York, New York. There are two baby grand piano's pushed together and the artists play for tips. The better you tip them, the faster your song gets played. Here's the fun part though, they will reward you if you request a song they cannot play. The experience is amazing, they play anything from Billy Joel's piano man, to Snoop Dog's no diggity.
Make sure you get there early seeing as the place fills up quite quickly. You can easily spend the whole night there singing and dancing your ass off. A VERY memorable time!

No one has mentioned "The Red Rooster" or "The Green Door" or "The Studio". The first two are swingers clubs, and the latter is a store dedicated to electro-stimulation. My girlfriend and I went there one time and there was a naked woman trying out a chair they build that has a fucking machine attached. Very interesting.

Oh I've come here just in time for you ....Las Vegas is called as the City of Sin....and one of the sexiest city in the country with some of the best shopping malls and of course casinos .... but don't miss these:
1. Luxor nightlife.
2. Bay beach...excellent wave pool, lazy river, swimming pools, and private cabanas ;)
3. Strip House at Planet Hollywood Resort.... you wont regret that :P
4. Pussycat Dolls Lounge at Caesars Palace.
